教室概要
心臓は、ヒトでは1秒間に約1回の速さで拍動し全身に酸素と栄養分を送り続けている生命に不可欠の臓器である。心臓の拍動を制御しているタンパク質として種々のイオン制御因子(イオンチャネルやイオン輸送体、それらの制御因子として働くCa2+センサータンパク質など)が存在する。これらタンパク質は心臓だけでなく脳?神経系にも高発現してその興奮性制御に寄与し、記憶や学習など高次機能の調節にも関わっている。これらの異常は不整脈や心不全、記憶障害などの様々な心疾患?神経疾患を引き起こすことが知られている(図1)。しかし、未だ生理機能や病態との関連が不明なもの、また病態時や未成熟期のみ発現が上昇するものなども存在し、興味深い課題が残されている。
薬理学教室では篮球比分直播2年より就任した西谷(中村)友重教授を中心に、これらイオン制御関連タンパク質の循環器系、神経系、代謝系における新規生理機能の解明、病態との関連およびそのメカニズムの解明、またそれに基づく薬物治療への応用を目指した研究を行っている。またこれらの研究に必要な実験系も整っている(図2)。

一方、薬理学講座では長年にわたり、痛みの制御機構、鎮痛薬の薬理作用、脳疾患の病態生理に関する研究を行ってきた。特に最近では、体性感覚(痛み?痒み)異常および精神疾患?神経変性疾患の分子基盤に着目した研究を展開している。これらの分野に関してもイオンシグナルは重要であり、また神経系に働く薬剤の心毒性などに関しても興味深い。今後はこれら複数の領域を組み合わせ、ヒトiPS細胞由来の心筋?神経細胞なども用いることにより、幅広い観点からヒト循環器疾患、痛みを含めた神経疾患、代謝疾患の病態分子基盤解明を目指した研究を展開していく。
講師の陳以珊はアフリカツメガエルをin vitro 発現系として用い、電気生理学的解析により神経?心筋のイオンチャネル?受容体の構造―活性相関、および薬物によるタンパク質機能の制御機構に関する研究を行っている(図3)。
安田助教は、病気によって臓器に細胞外マトリックスが過剰に蓄積する臓器線維症と、細胞外マトリックス分解産物に由来する生理活性物質マトリクリプチンに着目した研究に取り組んでいる。心不全や高血圧症といった心血管疾患を中心として、各種臓器の線維化病態とその臓器?病態に対するマトリクリプチンの機能解明、およびそれに基づく新規治療薬開発を目指した研究を行っている。これらの研究を行うにあたり、個体?組織?細胞レベルでの心機能解析も行っている(図4)。
納富講師は、イオン動態?力学的刺激の生体恒常性維持と疾患に及ぼす影響について研究を推進している。近年では、光遺伝学を硬組織に適応して、骨代謝疾患との関連を検討してきた。この方法は循環器疾患研究にも適用できる。分子生物学?形態学?電気生理学手法を融合させながら、多様な疾患とイオン動態?力学的刺激の関係を探索するとともに、そのメカニズムに基づいた薬物の開発につなげることを目指す(図5)。
教室では定期的に抄読会を行っている。担当者が最近発表された興味深い文献を紹介し、スタッフ全員での討論を通じて得られた最新の情報を、研究の発展に繋げることを目的としている。
教育概要
生体における各種薬物の作用について、【個体?細胞レベルでの作用機序】および【薬効に影響を及ぼす因子】に関する基礎的知識を提供する。疾病の治療?診断?予防に用いる薬物の薬理作用と有害作用を十分に理解し、応用できる医療人の育成を目標とした教育を行っている。
